Wikipedia (http://en.wikipedia.org/wiki/The_Pursuit_of_Happyness) offers
an explanation:

"Unwilling to be separated from his son, Chris picks Christopher up from
his daycare in Chinatown before Linda can. Chris complains about a sign
outside the daycare center misspelling "happiness" as "happyness", hence
the spelling of the film's title."

(Without seeing the film, it's hard to say, but it seems this might be a
metaphor for the difference between what is happiness and what one
thinks should be happiness.)
